Pharmachologic effect:

Baclofen is a muscle relaxant and antispastic agent that works by inhibiting synaptic transmission in the central nervous system, primarily at the spinal level, resulting in relief from muscle spasticity.

Indications:


  • Treatment of spasticity associated with multiple sclerosis
  • Spinal cord injuries or diseases

Contraindications:


  • Hypersensitivity to baclofen or any of the excipients
  • Peptic ulcer
  • Severe renal impairment

Side effects:


In order of decreasing probability: Drowsiness, Dizziness, Weakness, Fatigue, Headache, Insomnia, Nausea, Constipation, Confusion, Hypotension

Dosing and Administration:


The initial dose is usually 5 mg taken orally three times a day. The dose may be increased by 5 mg every three days until the desired effect is achieved. The usual maintenance dose ranges from 40 mg to 80 mg per day, divided into several doses. The maximum recommended dose is 80 mg per day.
