Pharmachologic effect:

Acyclovir is an antiviral agent that inhibits the replication of herpes simplex virus (HSV) types 1 and 2, varicella-zoster virus (VZV), Epstein-Barr virus (EBV), and cytomegalovirus (CMV). It acts by converting into acyclovir triphosphate, which inhibits viral DNA synthesis.

Indications:


  • Treatment of herpes simplex virus infections of the skin and mucous membranes, including initial and recurrent genital herpes.
  • Suppression of recurrent herpes simplex infections in immunocompetent patients.
  • Prophylaxis of herpes simplex infections in immunocompromised patients.
  • Treatment of varicella (chickenpox) and herpes zoster (shingles).

Contraindications:


  • Hypersensitivity to acyclovir or any component of the formulation.
  • Severe renal impairment without appropriate dose adjustment.

Side effects:


In order of decreasing probability: Nausea, Diarrhea, Headache, Dizziness, Fatigue, Abdominal pain, Rash, Itching, Vomiting

Dosing and Administration:


For the treatment of herpes simplex infections, the recommended dose is 200 mg taken five times a day for 5 days. For the suppression of recurrent infections, 400 mg twice daily is recommended. For the treatment of varicella and herpes zoster, 800 mg should be taken five times a day for 7 to 10 days. Dosage adjustments may be necessary for patients with renal impairment.
