Clopidogrel is an antiplatelet agent that inhibits platelet aggregation by selectively and irreversibly binding to the adenosine diphosphate (ADP) P2Y12 receptor on platelets, preventing ADP-mediated activation of the glycoprotein GPIIb/IIIa complex. This action reduces the risk of thrombotic events such as myocardial infarction and stroke.Indications:
- Prevention of atherothrombotic events in patients with myocardial infarction, ischemic stroke, or established peripheral arterial disease.
- Prevention of atherothrombotic and thromboembolic events in atrial fibrillation.
- Acute coronary syndrome (ACS) including non-ST segment elevation ACS and ST segment elevation myocardial infarction.
Contraindications:
- Hypersensitivity to clopidogrel or any component of the formulation.
- Active pathological bleeding such as peptic ulcer or intracranial hemorrhage.
- Severe hepatic impairment.
Side effects:
In order of decreasing probability: Common: Bleeding, bruising., Uncommon: Diarrhea, abdominal pain, dyspepsia., Rare: Thrombocytopenia, neutropenia, aplastic anemia., Very rare: Severe allergic reactions, including anaphylaxis.
Dosing and Administration:
The recommended dose of Clopidogrel Canon is 75 mg once daily, with or without food. In acute coronary syndrome, an initial loading dose of 300 mg may be given, followed by 75 mg once daily.
