Pharmachologic effect:

Alogliptin is a selective inhibitor of the enzyme dipeptidyl peptidase-4 (DPP-4), which slows the inactivation of incretin hormones, thereby increasing their levels in the bloodstream. This leads to an increase in insulin synthesis and release from pancreatic beta cells and a decrease in glucagon secretion from pancreatic alpha cells, resulting in improved blood glucose control.

Indications:


  • Type 2 diabetes mellitus to improve glycemic control in adults as an adjunct to diet and exercise.

Contraindications:


  • Hypersensitivity to alogliptin or any of the excipients
  • Type 1 diabetes mellitus
  • Diabetic ketoacidosis
  • Severe renal impairment
  • Severe hepatic impairment

Side effects:


In order of decreasing probability: Headache, Nasopharyngitis, Upper respiratory tract infection, Gastrointestinal disturbances, Rash, Hypoglycemia (when used with insulin or sulfonylureas), Pancreatitis

Dosing and Administration:


The recommended dose of Vipidia is 25 mg once daily, with or without food. Adjustments may be necessary based on renal function.
