Pharmachologic effect:

Amlodipine is a calcium channel blocker that dilates blood vessels and improves blood flow. It is primarily used to treat high blood pressure (hypertension) and chest pain (angina). By relaxing the blood vessels, it helps to reduce the workload on the heart and lower blood pressure.

Indications:

• Hypertension
• Chronic stable angina
• Vasospastic angina (Prinzmetal's or variant angina)

Contraindications:

• Severe hypotension
• Shock (including cardiogenic shock)
• Obstruction of the outflow tract of the left ventricle (e.g., high-grade aortic stenosis)
• Hemodynamically unstable heart failure after acute myocardial infarction
• Known hypersensitivity to amlodipine or any of the excipients

Side effects:

In order of decreasing probability: Very common: Edema, Common: Dizziness, headache, flushing, palpitations, abdominal pain, nausea, fatigue, Uncommon: Vomiting, dyspepsia, altered bowel habits, rash, muscle cramps, dyspnea, Rare: Confusion, mood changes, increased sweating, visual disturbances, Very rare: Hepatitis, jaundice, pancreatitis, Stevens-Johnson syndrome

Dosing and Administration:

The usual initial antihypertensive oral dose of amlodipine is 5 mg once daily, which may be increased to a maximum dose of 10 mg depending on the individual patient's response. For angina, the recommended dose is 5-10 mg once daily. Dosage should be adjusted according to the patient's needs and response.

Storage:

Store at a temperature not exceeding 25°C (77°F).
