Pharmachologic effect:

Metformin Sanofi is an oral antihyperglycemic drug used to manage type 2 diabetes. It works by decreasing hepatic glucose production, decreasing intestinal absorption of glucose, and improving insulin sensitivity by increasing peripheral glucose uptake and utilization.

Indications:


  • Type 2 diabetes mellitus, particularly in overweight patients, when dietary management and exercise alone do not result in adequate glycemic control.

Contraindications:


  • Hypersensitivity to metformin or any of the excipients
  • Acute or chronic metabolic acidosis, including diabetic ketoacidosis, with or without coma
  • Severe renal impairment (eGFR below 30 mL/min/1.73 m²)
  • Acute conditions with the potential to alter renal function such as dehydration, severe infection, shock
  • Acute or chronic disease which may cause tissue hypoxia (e.g., cardiac or respiratory failure, recent myocardial infarction, shock)
  • Hepatic insufficiency
  • Excessive alcohol intake, acute or chronic

Side effects:


In order of decreasing probability: Very common: Gastrointestinal disorders such as nausea, vomiting, diarrhea, abdominal pain, and loss of appetite, Common: Taste disturbance, Uncommon: Decreased vitamin B12 levels, Rare: Lactic acidosis, skin reactions such as erythema, pruritus, urticaria, Very rare: Hepatitis, liver function test abnormalities

Dosing and Administration:


The usual starting dose is 500 mg or 850 mg once daily, given during or after meals. The dose may be increased gradually to a maximum of 2000 mg daily, divided into 2-3 doses. Dose adjustments should be based on blood glucose measurements.

Storage:


Store at a temperature not exceeding 25°C (77°F).
