Pharmachologic effect:

Allopurinol is a xanthine oxidase inhibitor that reduces the production of uric acid in the body, helping to prevent gout attacks and kidney stones.

Indications:


  • Gout
  • Hyperuricemia
  • Recurrent calcium oxalate stones

Contraindications:


  • Hypersensitivity to allopurinol
  • Acute gout attack
  • Severe renal impairment

Side effects:


In order of decreasing probability: Rash, Nausea, Diarrhea, Drowsiness, Headache, Dizziness, Hypersensitivity reactions

Dosing and Administration:


The recommended starting dose is 100 mg per day, which may be increased by 100 mg weekly until the desired uric acid level is achieved. The typical maintenance dose ranges from 200 mg to 300 mg per day for mild conditions, and 400 mg to 600 mg per day for more severe conditions. It should be taken after meals with plenty of fluids.
