Pharmachologic effect:

Vildagliptin is an oral antihyperglycemic agent that works by inhibiting the enzyme dipeptidyl peptidase-4 (DPP-4), which results in increased levels of incretin hormones. This leads to enhanced insulin secretion and decreased glucagon levels, thereby improving glycemic control in patients with type 2 diabetes mellitus.

Indications:


  • Type 2 diabetes mellitus as monotherapy
  • Type 2 diabetes mellitus in combination with other oral antidiabetic agents
  • Type 2 diabetes mellitus in combination with insulin

Contraindications:


  • Hypersensitivity to vildagliptin or any of the excipients
  • Type 1 diabetes mellitus
  • Diabetic ketoacidosis
  • Severe renal impairment
  • Moderate to severe hepatic impairment

Side effects:


In order of decreasing probability: Common: Headache, dizziness, nausea, Uncommon: Hypoglycemia, fatigue, constipation, Rare: Hepatic dysfunction, skin lesions, pancreatitis

Dosing and Administration:


The recommended dose of vildagliptin is 50 mg once or twice daily. When used in combination with a sulfonylurea, the recommended dose is 50 mg once daily. The maximum daily dose is 100 mg. Vildagliptin can be taken with or without food.

Storage:


Store at a temperature not exceeding 30°C (86°F).
